Significance

Geldanamycin is a member of the ansamycin class of natural products and as such it is a potential antitumor agent. Furthermore, it is the first reported Hsp90 inhibitor, a protein regulating folding, transport and degradation of other proteins. The synthesis features two diastereo­selective Hosomi-Sakurai-type crotylations to set the C-10, C-11 and C-14 stereogenic centers.
