Syntheses of functionalized cyclooctynes for bio- and surface-orthogonal applications are described and compared. Points of interest are their chemoselective reactivity in cycloadditions and their compatibility with transannular side reactions. Applications of cyclooctynes for layer-by-layer-synthesis on the Si(001) surface are presented.
