A variety of acid-catalyzed conditions for oxazolidin-5-one formation from Fmoc protected amino acids and paraformaldehyde have been examined. Good yields of cyclized monosubstituted amino acids were obtained at room temperature with BF3 ⋅ OEt2 or PTSA/silica gel after short reaction times, or with PTSA/MgSO4 after extended periods. Cyclization of α,α-disubstituted amino acids is less successful.
