Significance

Hypercholesterolemia increases the risk of coronary heart disease and can be controlled by inhibiting the enzyme HMG-CoA reductase which blocks the rate-limiting step of cholesterol synthesis. An efficient second-generation synthesis of a pyrrole-based HMG-CoA reductase inhibitor I, which is a regioisomer of atorvastatin (Lipitor®), is reported here.
