An eight-step reaction sequence for simultaneous multiple synthesis of 1,2,6-trisubstituted 1,2,3,4-tetrahydroisoquinolines 17 on 2-hydroxyethyl polystyrene has been developed. Each target is comprised of a common fixed building block and four variable ones which are selected from three commercially available compound sets (ω-halogenated fatty acids, primary amines (2x), and acid chlorides). The assemblage of a prototype library featuring 24 discrete members serves to illustrate our protocol.
