Tris-bridged cyclic hexapeptide containing hydrophilic amino acid residues as an active site of lipase was synthesized from the piperazin-2-one derivatives which were obtained by treatment of the corresponding substituted dipeptides with p-toluenesulfonic acid or trifluoroacetic acid in toluene.
