Summary

  1. Visnagin was produced and isolated in crystalline form from A. visnaga tissue cultures The maximum visnagin concentration produced in Erlenmeyer cultures was 0.31 per cent, and in a New Brunswick fermentor was 0.13 per cent.

  2. β–carotene was present in the suspension cells in a concentration of 0.05 mg/100 gm wet weight.

  3. From the growth data of A. visnaga suspension cultures grown in Erlenmeyer flasks, the maximal visnagin concentration from cells (301 mg/100 gm dry wt) and from medium (11 mg/100 ml) occurred on the 7th day. The maximum cell growth (0.62 gm dry wt/50 ml medium) was on the 14th day.

  4. The maximum total visnagin production from eight 21 day fermentations was 142 mg and occurred in a fermentor receiving an aeration rate of 4,000 cc/min and agitation rate of 150 rpm (Fermentation IV–B). The maximum cell concentration (132.0 mg/100 gm dry wt) was obtained from two 35 day fermentations receiving an aeration rate of 1000 cc/min and agitation rate of 100 rpm (Fermentation V–G).

However, in another set of three fermentations (Fermentation VI), one 7 day fermentor produced a greater cell concentration of visnagin (58 mg/100 gm dry wt) than the other two 21 day fermentors. It therefore appears from this experiment, and from the Erlenmeyer growth study data, that visnagin production may be greater during the earlier stages of suspension cell growth.

Zusammenfassung

Aus Gewebekulturen von Ammi visnaga wurde Visnagin in kristalliner Form isoliert. Außerdem fand sich β–Carotin. Die höchste Konzentration trat am 7. Tage, das maximale Zellwachstum am 14. Tage auf.
