Significance

The key step in the multikilogram synthesis of the drug candidate is a diastereoselective asymmetric cyclopropanation involving insertion of a carbene derived from ethyl diazoacetate into the alkene C mediated by Nishiyama’s catalyst (Tetrahedron: Asymmetry 1995, 6, 2487).
