Significance

ZD6126, an anticancer phosphate prodrug of N-acetylcolchinol, is under development by AstraZeneca. This synthesis was accomplished in seven steps and the key step involves asymmetric enamide hydrogenation in the presence of a ruthenium catalyst (see also: Tetrahedron Lett. 2007, 48, 4623).
