Significance

The authors reported the synthesis of a nucleoside containing a rigid nitroxide spin lab (1) via a convergent synthetic strategy. The phen­oxazine functionality of 1 was aimed to specifically bind guanine through hydrogen bonds. EPR spectroscopic studies on this spin-labeled nucleoside, spin-labeled oligomer, and spin-labeled DNA duplex clearly demonstrated the limited mobility of spin label within a DNA duplex, indicating its potential application for DNA structural analysis. In addition, compound 1 was found to be easily reduced into fluorophores 2, which are attractive probes for biophysical studies of DNA.
