Zusammenfassung

DFP32 wird nach intramuskulärer und intravenöser Injektion sehr rasch in die Kaninchenerythrozyten aufgenommen, der Markierungsvorgang ist nach spätestens 2 Std. beendet. Der Anteil des an die Erythrozyten gebundenen Stoffes steigt mit Verringerung der injizierten Dosis an, ebenso mit Verlängerung der Infusionsdauer bei intravenöser Applikation. In den ersten 24 Std. nach Markierung wird ein deutlicher „Frühabfall” der Erythrozytenradioaktivität beobachtet, der am ehesten durch die partielle Elution des Markierungsmittels in diesem Zeitraum bedingt ist. Bei Vernachlässigung der ersten 24 Std. eignet sich DFP32 gut zur Bestimmmung der Lebenszeit der Kaninchenerythrozyten, wobei der Verlauf der Abfallskurve Rückschlüsse auf die Kinetik des Erythrozytenabbaues zuläßt. Der Verlauf der Erythrozytenradioaktivität nach gleichzeitiger Markierung mit DFP32 (in vivo) und Cr51 (in vitro) weist darauf hin, daß eine stetige Elution des Cr51 aus den markierten Zellen in der Größenordnung von 1% am Tag stattfindet, wie sie auch beim Menschen zu beobachten ist.

Summary

Following intramuscular and intravenous injection, DFP32 is very rapidly taken up by rabbit erythrocytes. Labelling is completed within 2 hours at the latest. The fraction of the label bound to red cells increases with decreasing amounts injected and also with prolongation of the infusion when given intravenously. During the first 24 hours following labelling a significant early loss of red cell radioactivity can be observed and this is most likely due to partial elution of the label during this period. If the results obtained during the first 24 hours are discarded DFP32 appears to be useful for determining the life span of rabbit erythrocytes, the form of the curve allowing conclusions to be drawn as to the kinetics of red cell destruction. The results obtained from simultaneous labelling with DFP32 (in vivo) and Cr51 (in vitro) indicate that there is a continuous elution of Cr51 from the labelled cells of the order of 1% per day, similar to the one observed in the human subject.

Résumé

DFP32 est incorporé rapidement dans les globules rouges des lapins après injection intramusculaire ou intraveneuse. Le marquage est terminé au plus tard après deux heures. Le pourcentage de la substance fixée dans les globules rouges monte quand la dose injectée est diminuée. Des variations de la durée de l’infusion intraveneuse produit également un effet sur le pourcentage de fixation. Dans les premières 24 heures après le marquage on observe une décroissance préliminaire nette qui s’éxplique par une élution partielle de la radioactivité dans cette période. Quand on renonce à les premières 24 heures DFP32 est bien utilisable pour la détermination de la période de vie des globules rouges des lapins. La forme de la courbe de décroissance permet des conclusions sur la cinétique de la destruction des globules rouges. Une comparaison de la radioactivité des érythrocytes marqués simultanément par le DFP32 in vivo et par le chrome51 in vitro donne évidence d’une élution permanente du chrome51 des globules marqués dans l’ordre de 1% par jour comme on l’observe également chez l’homme.
