Significance

Modular synthetic approach to asymmetric-structured near-infrared (NIR) fluorophores 1 is described. The key to the stepwise synthesis is a successful isolation of intermediate (Int-A) in the first condensation of diketopyrrolopyrrole (DPP). The validity of this method is also demonstrated by synthesis and internalization of the cell-penetrating, oligopeptide-conjugated fluorophore 2 in living cells.
