Significance
Prostaglandin E2 is a proinflammatory mediator that exerts its physiological action by binding to the G-protein coupled receptor EP1. GSK269984B is an EP1 antagonist that was a clinical candidate for the treatment of inflammatory pain. The key step in the synthesis depicted is a magnesium-promoted phenolic aldol reaction of the 2-formylpyridine B (M. Whiting, M. C. Wilkinson, K. Harwood Synlett 2009, 1609).
