Direct Solid-Phase Synthesis of the β-Amyloid (1-42) Peptide

Significance

A microwave-assisted solid-phase synthesis of the 42-mer β-amyloid peptide was described. The coupling of Fmoc amino acids (Fmoc AA) with peptide chains attached to a ­ChemMatrix resin (conditions A) followed by deprotection of the Fmoc group under microwave conditions gave the corresponding resin-supported AA peptides. The coupling of Fmoc-His(Trt)-OH was performed at room temperature without microwave irradiation in order to avoid the racemization (conditions B). The desired β-amyloid peptide was obtained via repetition of the protocol within 15 hours overall processing time in 78% crude yield and 83% crude purity.
