Diskussionsbeitrag zur Bedeutung der nichttrizyklischen Antidepressiva für die Theorienbildung der Depression

Abstract

Clinical and experimental research on antidepressive drugs during the 10 years which elapsed since Kuhn's first paper in 1957 centered mainly around the tricyclic compounds. This gave considerable support to the noradrenalin hypothesis of depression, although it did not furnish any definite proof. Since 1968, reports have been published on the antidepressive action of non-tricyclic compounds, stimulating the exploration of new theoretical avenues. The article discusses briefly the theoretical prerequisite of ,,emotional integration" as an important factor in depression and of the ,,double receptors" for parkinsonism, using the development of trazodone as an example.

Zusammenfassung

Zehn Jahre Entwicklung antidepressiver Substanzen seit der Arbeit von Kuhn 1957 galt vorwiegend trizyklischen Verbindungen. Dadurch wurde die Nor-Adrenalin-Hypothese der Depression sehr gestützt, wenn auch nicht bewiesen. Die Beobachtung der antidepressiven Wirkung von nicht-trizyklischen Verbindungen seit 1968 gibt Anlaß zu neuen theoretischen Überlegungen. Am Beispiel der Entwicklung von Trazodone wird die theoretische Voraussetzung der ,,emotionalen Integration" für die Depression und der ,,doppelten Rezeptoren" für den Parkinsonismus kurz diskutiert.
