Validated HPLC-DAD method for resveratrol determination during identification in raw materials and stability, preformulation, permeability studies

Resveratrol (3,4′,5- trihydroxystilbene) is a nutraceutical that has exerted anti-inflammatory and antioxidant activity [1]. It has been found also as anticancer agent, and substance with effect on neurological diseases and other maladies[2].
The aim of this study was to develop an HPLC-DAD method suitable for resveratrol determination during identification in raw materials and its stability, preformulation and permeability studies.
The HPLC method with UV detection with the Kinetex C18 column (100 × 2.1 mm, 5 μm) was used. The mobile phase consisted of ACN/0.5% acetic acid in ration 20/80. The detection wavelength was 306 nm, the column temperature was set at 40°C. The method meets all required validation parameters (selectivity, linearity, precision, accuracy). The changes of resveratrol permeability was investigated by using PAMPA test.
The proposed RP-HPLC method is a suitable technique for the determination of resveratrol in different approaches such as preformulation, stability and permeability studies. Resveratrol permeability coefficient (pH 7.4) was 74.89±27.28x10-6 cm/s, what classified drug as well-permeable.
