Genkwa Flos is the flower buds of Daphne genkwa Sieb. et Zucc. (Thymelaeaceae)a traditional medicine in Korea and China [1], [2]. enkwanin is characterized as the reference phytochemical for the quality control of this herbal medicine in Chinses and Taiwan Pharmacopoeia. ive flavonoids including genkwanin in the Genkwa Flos using ultra-performance liquid chromatography coupled with a photodiode array detector. o optimize the condition of extraction, which mapped out a mixture matrix design (MMD) for extract solvent and three-level full factorial design with regression analysis [3]. Water, methanol, and acetone were as the extraction solvents for MMD and the extraction method. Following multiple regression analyses of, 3D response surface and the contour plots to determine the optimal conditions. extractsolvent44% water, 56% acetone, and 0% methanol12 hours150 rpm70℃. This work proposed a method for the optimal extraction of flavonoids in Genkwa Flos and the determination of five flavonoids apigenin and genkwanin by UPLC-DAD.
