Summary
By treating rats with lithium chloride or cocaine hydrochloride, or lithium chloride followed by cocaine hydrochloride, we have shown the angatonistic effects of these drugs on two mechanisms that may be involved in regulating serotonin 5-HT) synthesis in the striate cortex. Lithium chloride (5 to 10 meq/kg/day) stimulates the relative velocity of the active uptake of labelled tryptophan and proportionally enhances the conversion of labelled tryptophan to 5-HT in synaptosomally enriched preparations. With continued administration of lithium chloride, the activity of tryptophan hydroxylase from the median raphe and subsequently in lysed synaptosomal preparations from striate cortex is reduced; the substrate uptake remains enhanced, but the conversion of substrate to transmitter returns to control levels. In contrast, an injection of cocaine hydrochloride inhibits the high affinity uptake of tryptophan, reducing the conversion of the amino acid to 5-HT and resulting in an increase in the biosynthetic enzyme activity. However, administration of cocaine hydrochloride (100 mg/kg) after three daily lithium chloride injections (10 meq/kg) results in no apparent effects on substrate uptake, conversion, or enzyme activity. We theorize that the effect of lithium was to push two regulatory parameters (the uptake of substrate and the enzyme activity) to their respective functional upper and lower limits, leaving the serotonergic neurons “buffered” against the “usual” effects of the stimulant drug, and offer this neurobiological model for consideration in relation to the clinical effects of lithium in the prophylaxis of both mania and depression in some patients.

Zusammenfassung
Wir haben Ratten mit Lithiumchlorid oder Cocain-hydrochlorid und mit Lithiumchlorid und nachher Cocain-hydrochlorid behandelt und den antagonistischen Effekt dieser Drogen auf zwei Mechanismen aufgezeigt, die in der Regulation der Serotonin (3-HT) Synthese in der Rinde des Striatum beteiligt sein können. Lithiumchlorid (3–10 meq/mg/am Tag) erhöht die relative Geschwindigkeit der aktiven Aufnahme markierten Tryptophans und steigert im Verhältnis die Wandlung markierten Tryptophans zu 3-HT in synaptosomal bereicherten Präparaten. Wenn man mehr Lithiumchlorid gibt, wird die Aktivität der Tryptophanhydroxylase von der Mittel-Linie und danach in lysierten synaptosomalen Präparaten der Rinde des Striatum gemindert. Die Aufnahme des Substrats bleibt erhöht, aber die Wandlung des Substrats zu dem Transmitter kehrt zu dem Niveau der Kontrollen zurück. Wenn man dagegen Cocain hydrochlorid injiziert, hindert es die hochgradige Affinitäts-Aufnahme des Tryptophan, vermindert die Wandlung der Aminosäure zu 3-HT. Das Resultat ist ein Anstieg in der Aktivität biosynthetischer Enzyme. Wenn man aber Cocain hydrochlorid (100 mg/kg) nach 3 täglichen Injektionen von Lithiumchlorid (10 mg/kg) gibt, dann findet man keine sichtbaren Folgen für die Aufnahme des Substrats, Konversion oder Enzym-Aktivität. Wir nehmen theoretisch an, daß der Lithium-Effekt zwei regulatorische Parameter (die Aufnahme des Substrats und die Enzymaktivität) auf ihre jeweiligen maximalen und minimalen Grenzen trieb und die serotonergischen Neurone gegen die “übliche” Wirkung der Reizdroge abschirmte. Wir bieten dieses neurobiologische Modell an als einen Beitrag zur klinischen Wirkung von Lithium in der Prophylaxe von sowohl Manie wie Depression bei gewissen Patienten.
