Significance

Cytotrienin A is an antitumor ­secondary metabolite isolated from the fermentation broth of the soil bacteria Streptomyces sp. RK95-74. The E,E,E-trienine moiety within a 21-membered cyclic lactam is the common structural feature of all ansamycins. Herein, the first total synthesis is reported featuring a late-stage macrocyclization using ring-closing metathesis.
