Significance

The concise synthesis of SNAP-7941, a chiral dihydropyrimidone is reported. The procedure involves an organocatalytic key step to optically pure intermediate 1. Two approaches, a cinchona alkaloid catalyzed Mannich reaction and a chiral phosphoric acid catalyzed Biginelli reaction proved to be highly useful for this synthesis.
