Significance

A synthesis of the hepatitis C virus protease inhibitor BILN 2061 features detailed optimization of a ring-closing metathesis (RCM) to construct the 15-membered ring. The route described delivered over 100 kg of G and the authors claim this is probably the first large-scale application of the RCM reaction. However, the experimental only describes the reaction on a 23.8-mmol scale.
