Significance

The authors examined the regio­selectivity and reactivity of several halogenated purines in cross-coupling reactions. Employing the N-benzyl-protected purine (1) as a building block, they successfully synthesized a cyclic tetrameric purine-palladium complex (8), via repetitive transmetallation, iodo-de-chlorination and cross-coupling reactions. This stepwise synthetic route afforded 8 in eight steps and 17% overall yield.
