Significance

NS9544 acetate is a monoamine reuptake inhibitor that is a candidate for the treatment of neuropathic pain. After all attempts at a classical resolution of intermediate E failed, an asymmetric synthesis was undertaken in which the chirality was introduced by asymmetric deprotonation of tropinone A using the chiral lithium amide base B.
