Zusammenfassung

Studienziel: Ziel der Studie war es, den Einfluss von Dexamethason im Nährmedium von humanen stromalen Zellen aus dem Knochenmark (human bone marrow stromal cells, hBMSC) und zyklischem, mechanischem Dehnungsstress auf die osteogene Differenzierung, quantifiziert durch die Bestimmung der mRNA-Expression von Kollagen Typ I, II, III, Tenascin C und Cbfa1 zu untersuchen. Methode: hBMSCs von sieben Spendern mit einem Alter von 32,5 ± 6,2 Jahren wurden ab der ersten Passage mit (D +) oder ohne Dexamethason (D –) kultiviert. Nach der zweiten Passage wurden je 2,2 × 105 Zellen in flexible Silikonschalen überführt. Nach ihrer Adhäsion auf dem Schalengrund wurden die Zellen einem Dehnungsstress unterzogen von 2 % (D + 2; D – 2), 8 % Dehnung (D + 8; D – 8) und einer Kontrollgruppe ohne Stress (D + 0; D – 0). Der Stress wurde an drei aufeinander folgenden Tagen mit einer Frequenz von 1 Hz und täglich drei Zyklen Stress mit einer Dauer von jeweils zwei Stunden durchgeführt. Die Zellproben wurden am ersten Stresstag, am ersten Tag nach Stress (Tag 4) und am vierten Tag nach Stress (Tag 7) entnommen. Die Quantifizierung der Genexpression von Kollagen Typ I, II und III, Tenascin C und Cbfa1 erfolgte durch RT‐PCR. Ergebnisse: Die mRNA-Expression von Cbfa1 zeigte an den unterschiedlichen Tagen signifikante Unterschiede (p < 0,05) sowie für die Dehnung an Tag 1: 8 %: 0,0291 ± 0,0338 versus 0 %: 0,00 528 ± 0,0127, p = 0,017; Tag 8: 8 %: 0,0411 ± 0,116, 0 %: 0,00 103 ± 0,00 217, p = 0,009. Die Beobachtung der anderen Parameter zeigte zwar Tendenzen auf, erbrachte jedoch keine statistisch signifikanten Unterschiede. Schlussfolgerung: Die Arbeit hat gezeigt, dass über den kurzen Beobachtungszeitraum von sieben Tagen zyklischer mechanischer Dehnungsstress ein stärkerer osteogener Differenzierungsinduktor ist als Dexamethason.

Abstract

Aim: The aim of the study was to investigate the effects of dexamethasone and cyclic mechanical strain on human bone marrow stromal cells (hBMSCs) in osteogenic differentiation by determing levels of mRNA of collagen I, II, III, tenascin C and Cbfa1. Method: hBMSCs from seven donors (32.5 ± 6.2 years old) were cultivated with (D +) or without (D –) dexamethasone. After the second passage 2.2 × 105 cells were seeded on flexible silicon dishes. A cyclic mechanical strain with an elongation of 2 % (D + 2; D – 2) or 8 % (D + 8; D – 8) was applied for three days with a stimulation time of three times for two hours each day. Cells were harvested on day 1, day 1 after stress (day 4) and day 4 after stress (day 7). mRNA expression of collagen I, II, III, tenascin C and Cbfa1 was investigated by RT‐PCR. Results: Cbfa1 mRNA levels were significantly different on different days (p < 0.05), and for strain on day 1: 8 %: 0.0291 ± 0.0338 versus 0 %: 0.00 528 ± 0.0127, p = 0.017; day 8: 8 %: 0.0411 ± 0.116, 0 %: 0.00 103 ± 0.00 217, p = 0.009. All other observed paramaters showed tendencies without significant differences. Conclusion: In the short-term over seven days, cyclic stretching is a stronger differentiation factor than dexamethasone.
