Cytokine Profiles of Autologous Conditioned Serum (ACS) and Autologous Protein Solution (APS) from horses with Pituitary Pars Intermedia Dysfunction (PPID) and Unaffected Controls

Introduction: Middle-aged horses are commonly affected by joint disease, and within this population of horses, endocrine disease, such as PPID, is more prevalent. PPID is known to affect immune function. While orthobiologics such as ACS and APS have been used in this group, there is a gap in knowledge on how PPID affects the anti-inflammatory nature of these products. The main hypothesis was that horses with PPID would have altered cytokine profiles in ACS and APS compared with non-PPID horses.
Materials and Methods: Six PPID and six non-PPID horses from a single farm had blood drawn aseptically and processed for ACS and APS per the manufacturer’s directions. Aliquots of each ACS and APS preparation were frozen and stored at -80°C until batch analysis. Pro- and anti-inflammatory cytokine concentrations (IL-1β, IL-1Ra, IL-4, IL-6, IL-10, TNF-α, TGF-β, IGF-1) were determined in triplicate using commercial ELISA kits. Groups were compared with Wilcoxon signed-rank tests (p < 0.05).
Results: PPID horses were significantly older (median: 20 years) than non-PPID horses (median: 15 years) (p = 0.01). For both ACS and APS, there was no statistically significant difference in any of the cytokines when comparing PPID and non-PPID horses (0.08 > P > 1.00). When combining all horses, there was a significantly higher concentration of TFG-β in APS (median: 8,344 pg/mL) compared with ACS (median: 4,617 pg/mL) (p = 0.002).
Discussion/Conclusion: These data suggest PPID does not significantly change the cytokine profile of ACS and APS preparations tested. These results support further investigation of orthobiologics in aged horses with PPID.
