Phytohustil and root extract of marshmallow Althaea officinalis L. exert anti-inflammatory and anti-oxidative properties and improve the migratory capacity of HUVECs tested in an in vitro model of angiogenesis

    Introduction Phytohustil cough syrup (Phyto) containing root extract of Althaea officinalis L. (REA) is used for the treatment of lesioned laryngopharyngeal mucosa caused by dry coughs. Neovascularization at the site of the injury is central for wound healing. In this regard, anti-inflammatory/-oxidative phytochemicals that improve angiogenesis in the wound are essential to promote healing.

    Aim Our aim was to investigate the anti-inflammatory, anti-oxidative and pro-migratory properties of Phyto and REA on human umbilical vein endothelial cells (HUVEC).

    Method Treatment (24 h) with Phyto or REA, followed by 3 h lipopolysaccharide (LPS, pro-inflammatory) or 1 h H2O2 (pro-oxidative). Intracellular reactive oxygen species (ROS) were quantified with dichlorofluorescin (DCFDA) and interleukin- (IL-) 6 release (enzyme-linked immunosorbent assay, ELISA). The effect on the migratory capacity of HUVECs was determined by scratch assay.

    Results 100 µg/ml Phyto or REA inhibited the IL-6 release by 17% and by 16% compared to 0.1 µg/ml LPS. 500 µg/ml Phyto, its excipients or 500 µg/ml REA inhibited the IL-6 release by 17%, 24% or 22% compared to 0.1 µg/ml LPS. After 24 h with Phyto or REA and additionally (60 min) with 200 µM H2O2, Phyto inhibited the ROS production by 18% (500 µg/ml) or by 16% (1000 µg/ml) as well as after REA by 33% (1000 µg/ml) compared to H2O2. After 6 h treatment, 500 or 1000 µg/ml Phyto stimulated the wound closure by 10% and 1000 µg/ml REA by 13.5 % compared to control.

    Discussion Phyto has anti-inflammatory properties, protects against oxidative stress and improves the migratory capacity of HUVECs used as an in vitro model of the vascular endothelium. These properties of Phyto, observed with HUVECs may support the therapeutical effects of Phyto and may be also beneficial for patients during the treatment of laryngopharyngeal irritated mucosal membranes and, thus, may be appropriate for treatment of dry cough.
