Abstract
In vivo activity of an artificial surfactant was studied in surfactant depleted rats. After tenfold alveolar lavage, PaO2, tidal volume, and compliance of the respiratory system fell to one third of initial value. Substitution of large unilamellar vesicles containing 90 % Dipalmitoylphosphatidylchohne and 10 % unsaturated phosphatidylglycerol largely restored oxygenation and lung mechanics in most animals. Complete normalization with weaning from the ventilator, however, was achieved neither with liposomes nor with natural surfactant concentrate.
Zusammenfassung
Die Surfactant-deprivierte Ratte wurde zur Beurteilung der in vivo-Wirksamkeit eines künstlichen Surfactant-Substitutes verwendet. Nach zehnfacher alveolärer Lavage sanken Sauerstoffpartialdruck, Atemzugvolumen und Compliance des respiratorischen Systems auf ein Drittel des Ausgangswertes ab. Substitution von großen unilamellären Vesikeln aus 90% Dipalmitoylphosphatidylcholin und 10 % ungesättigtem Phosphatidylglycerol führte bei den meisten Tieren zu weitgehender Restitution von Oxygenierung und Lungenmechanik. Eine vollständige Normalisierung mit Entwöhnbarkeit vom Respirator konnte jedoch weder mit Liposomen noch mit natürlichem Surfactant-Konzentrat erzielt werden.
