Abstract

From extracts of roots of Daphnopsis racemosa GRISEB. (Thymelaeaceae) by combination of countercurrent distribution and chromatographic methods prostratin and seven irritant Daphnopsis factors R1-R7 were isolated. Spectroscopic characterization revealed presence of four known and four hitherto unknown tigliane, daphnane and 1α-alkyldaphnane type structures, respectively. As tigliane types the known prostratin (Ia) as well as the new 5β-hydroxy derivative thereof, Daphnopsis factor R2 (Ib), were identified. As daphnane types the known simplexin (Daphnopsis factor R3, IIa) and the so far unknown 5-deoxy derivative, Daphnopsis factor R4 (IIc), were isolated. Furthermore, the new Daphnopsis factors R6 and R7 of the 1α-alkyldaphnane type were identified as 5-deoxy derivatives IVc, IVd of Daphnopsis factors R1 and R5 (IVa, IVb). The latter are identical with the known Pimelea factors P2 and P3 isolated from Pimelea prostrata (Thymelaeaceae). - As may be seen from the irritant doses 50 and from the results of the assay for tumor promoting activity, presence of a 5β-hydroxy group in the tigliane, daphnane and 1α-alkyldaphnane type esters increases the biological activity as compared to the corresponding 5-deoxy-compounds.
