Zusammenfassung
Der Einfluß des Antispastikums Dantrolen auf die isometrische Kontraktion des M. adductor
pollicis wurde bei 14 Probanden unter chronischer Medikamenteneinnahme untersucht.
Neben einer dosisabhängigen deutlichen Reduktion der maximalen Kraftentwicklung fand
sich eine leichte Abnahme der Kontraktionszeit; die Relaxationszeit war demgegenüber
eher verlängert. Nach Absetzen der Medikation wurde bezüglich sämtlicher Parameter
eine teilweise zunächst überschießende Normalisierung beobachtet. Relativ häufig wurde
über Nebenwirkungen geklagt.
Die Befunde belegen den vorwiegend peripheren Angriffspunkt des Medikaments.
Summary
The effect of the muscle relaxant dantrolene sodium on the isometric contraction of
the adductor pollicis muscle was studied in 14 healthy volunteers during chronic medication.
Dose-related, a marked reduction of the maximal contraction force was observed. The
contraction time was slightly shortened, the relaxation time rather prolonged. Following
discontinuation of the medication, a partially overshooting normalization was found.
Side effects were observed quite often.
The findings support the predominant peripheral site of action of the drug.
