Summary
The efficacy and tolerability of vigabatrin were investigated in 55 adult patients with therapy-refractory symptomatic focal epilepsies. Fifteen patients were included in a single blind study (group I) and 40 patients were treated in an open uncontrolled study (group II). After two years 20 % of the patients in group I were still on vigabatrin, whereas the percentage after 2.4 years was 47.5 % in group II (reduction of seizure frequency of at least 50 %). Side effects were usually mild and transient. The possibility of provocation of generalized tonic clonic seizures and of psychoses has to be mentioned.
Zusammenfassung
Vigabatrin wurde zur Überprüfung des Therapieeffektes und der Nebenwirkungen bei insgesamt 55 Patienten mit symptomatischen fokalen Epilepsien eingesetzt (Gruppe I: Einfachblindstudie, n=15; Gruppe II: offene, unkontrollierte Studie, n=40). Nach 6 Jahren wurden in der Gruppe I noch 20 %, nach 2,4 Jahren in der Gruppe II noch 47,5 % der Patienten behandelt (Anfallsreduktion um mindestens 50%). Nebenwirkungen waren meist schwach ausgeprägt und passager. Auf die Möglichkeit einer Provokation von generalisierten tonisch-klonischen Anfällen und Psychosen ist hinzuweisen.
