Summary

In 70 children (including 47 infants) with severe, drug-resistant infantile spasms (West syndrome) vigabatrin led to a complete suppression of spasms in 29 patients and a more than 50 % reduction of seizure frequency in 46. This effect was most pronounced in patients with tuberous sclerosis and longterm efficacy was maintained in about 75 % of the patients with symptomatic infantile spasms. Tolerability of vigabatrin was evaluated in 136 children with observation periods up to 3 years. In about 30 % of patients adverse events were observed, the most common beeing agitation and muscular hypotonia.

Zusammenfassung

Von 70 Kindern (einschließlich 47 Säuglingen) mit schweren, therapieresistenten infantilen Spasmen (West-Syndrom) konnte unter Vigabatrin bei 46 eine mindestens 50%ige Besserung der Anfallsfrequenz beobachtet werden, und bei 29 kam es zu einer vollständigen Anfallsfreiheit. Dieser Effekt war bei Patienten mit symptomatischen infantilen Spasmen im Langzeitverlauf an. Die Verträglichkeit von Vigabatrin konnte bei 136 Kindern mit tuberöser Sklerose am ausgeprägtesten, und hielt bei rund 75% der Patienten mit einem Beobachtungszeitraum bis zu 3 Jahren bewertet werden. Bei rund 30% der Patienten traten Nebenwirkungen auf, am häufigsten Übererregbarkeit und muskuläre Hypotonie.
