Abstract
The influence of paroxetine (1 ×20mg/day) on safety-relevant performance was compared
with the effects of doxepin (2×50mg/day) and placebo. The medication covered a 3-week
period. On day 20 of treatment, ethanol was additionally administered (0.05% BAC).
The study group comprised 60 healthy male and female volunteers in the age range 37-60
years, who were assigned to the three structurally identical medication groups under
randomized, double-blind conditions. The functional capacity of primary interest was
investigated in seven tests to record visual orientation, forced concentration, simple
reaction time, choice reaction time, reaction under stress, vigilance, and motor co-ordination.
Test sessions took place before the treatment and five times during the medication
phase. Paroxetine proved comparable to placebo in all cases, while in comparison with
the two reference substances doxepin revealed loss of vigilance and motor coordination,
as well as of concentration and the simple (acoustic) reaction time.
Zusammenfassung
Der Einfluß von Paroxetin (1 ×20mg/d) auf sicherheitsrelevante Leistungen von Probanden
wurde verglichen mit den Wirkungen von Doxepin (2×50mg/d) und Plazebo. Die Medikation
ging über 3 Wochen. Am 20. Tag der Behandlung wurde zusätzlich Alkohol verabreicht
(0,5 % BAK). Die Studiengruppe bestand aus 60 gesunden, freiwilligen Männern und Frauen
im Alter von 37-60 Jahren, die randomisiert und doppelblind drei strukturgleichen
Medikationsgruppen zugewiesen wurden. Das primär interessierende Leistungsvermögen
wurde operationaüsiert durch sieben Tests zur Erfassung von optischer Orientierung,
Dauerkonzentration, Reaktion auf einfache Signale, Auswahlsignale und bei induziertem
Streßzustand, von Vigilanz und motorischer Koordination. Vor Behandlung und 5mal innerhalb
der Medikationsphase fanden Testsitzungen statt. Paroxetin erwies sich in allen Fällen
Plazebo vergleichbar, Doxepin zeigte gegenüber den beiden Vergleichs- substanzen Einbußen
bei Vigilanz und motorischer Koordination, Konzentration und akustischer Reaktionszeit.
