Significance

The synthesis of Amphidinolide A is a remarkable exemplification of the power of Ru-catalyzed alkene-alkyne coupling for the synthesis of 1,4-dienes. The reaction was used in the final step of the synthesis on unprotected substrate F to construct the C29 methylene and the 20 membered ring simultaneously.
