Concentration of nuclear receptors to oestradiolum in tumours chiasma-sellary (hypothalamo-pituitary) regions
Introduction: Receptors of sexual hormones, in particular to oestradiolum, are found out in various concentration in many areas of the central nervous system. It is proved, that sexual steroids and their receptors participate in various neuroendocrine reactions, during a differentiation of the central nervous system. It has led to occurrence of a hypothesis, that sexual steroids, in particular estrogens, concern to the factors participating in pathogenic and progressive growth of some tumors of a brain, and, that, probably, some tumors are hormone dependent. However, the mechanism of influence of sexual hormones on pathogenic tumors, in particular, adenomas of a hypophysis and tumors parasellary areas (cranyopharyngiomas, germinomas, chordomas and other rare tumors) finally is not revealed. Purpose: Definition of concentration of nuclear receptors to oestradiolum (ER) in various tumors of a hypophysis (prolactinumsecretory adenomas of a hypophysis, clinically-hormone inactive adenomas of a hypophysis – the CHIAH) and other tumours chiasma-sellary areas (cranyopharyngioma). Materials and methods: The material is received at operation from patients with prolactinomas (n=13), the CHIAH (n=31) and cranyopharyngiomas (n=20). A tumoral fabric homogenised, centrifuged. Nucleus allocated on Chauveau method updated by Kedrova and Orlova. At definition of amount of places of tumoral fabrics specifically connected in nuclear fraction used Anderson method based on an exchange marked of a hormone with unmarked. Amount of connecting places in nucleus expected in fetomole for 1mg of DNA, and the level of DNA – on method Burton. Results: At the analysis of the received data patients in groups with various tumors have been divided into two subgroups depending on concentration ER: with concentration ER ≤100 fmole/l DNA И>100 fmole/l DNA. In the second group has come: prolactinomas –11 cases from 13, the CHIAH –12 from 31, cranyopharyngiomas –7 from 20. Distinction is authentic between group prolactinumsecretory adenomas and as the CHIAH (p<0,05, by criterion xИ2), and cranyopharyngiomas (p<0,05, by criterion xИ2). Conclusion: Concentration of nuclear receptors to oestradiolum is higher in prolactinumsecretory adenomas of a hypophysis in comparison with clinically-hormone inactive adenomas of a hypophysis and cranyopharyngiomas.
