Summary

Rats were given 125I-labelled iodochloroxyquinoline (ICOQ, Vioform®, CIBA) orally. Radiochromatograms of thyroid hydrolysates 8, 24, 48 and 72 hrs after administration of the drug revealed that the 125iodide was transferred almost quantitatively from ICOQ to the thyroidal iodo-amino acids within 24 hrs. It is thus possible that the iodine contained in the ICOQ molecule may be actively utilized for the formation of thyroid hormones although the exact nature of the reaction has not been definitely clarified. ICOQ is not deiodinated extrathyroidally.

Résumé

125I-iodochloroxyquinoline (ICOQ, Vioforme®, CIBA) fut administré aux rats. Des radiochromatogrammes faits des hydrolysates thyroïdiennes 8, 24, 48 et 72 heures après l’administration de ce médicament revélèrent que l’iode-125 était presque complètement transféré de l’ICOQ aux acides aminés thyroïdiens dans 24 heures. Il semble que l’iode contenu dans la molécule ICOQ soit activement utilisé pour la formation des hormones thyroïdiennes bien que la nature exacte de cette réaction n’ait pas été finalement clarifiée. L’ICOQ n’est pas deiodé au dehors de la thyroïde.

Zusammenfassung

125J-markiertes Jodchlor-Oxychinolin (ICOQ Vioform® CIBA) wurde peroral an Ratten verabreicht. Die Radiochromatographie des Schilddrüsengewebes wurde nach enzymatischer Hydrolyse 8, 24, 48 und 72 Std. nach Verabreichung der Dosen durchgeführt. Es zeigte sich dabei, daß das 125J des ICOQ innerhalb von 24 Std. beinahe quantitativ zu den Jod-Aminosäuren überführt war. Es könnte deshalb möglich sein, daß das Jod des ICOQ in der Schilddrüse für die Synthese der Schilddrüsenhormone verwendet wird. Die exakte Reaktion, die für die Überführung des Jodes verantwortlich ist, wurde in diesen Versuchen nicht abgeklärt. ICOQ wird nicht extrathyreoidal dejodiert.
