Pregnane glycosides from Asclepiadoideae stimulate glucagon-like peptide-1 secretion in intestinal neuroendocrine cells

In the search for natural glucagon-like peptide-1 (GLP-1) secretagogues, which could act as antidiabetic compounds [1], an ethanolic extract from the Malagasy Cynanchum marnierianum (Apocynaceae, Asclepiadoideae subfamily) was found to significantly stimulate GLP-1 secretion in the intestinal secretin tumor cell line (STC-1). The bioguided fractionation led to the isolation of two new pregnane glycosides named marnieranosides A and B. They present structural analogies with pregnanes previously described in some Asclepiadoideae species known for their appetite suppressant and antihyperglycemic effects, such as P57 from Hoodia gordonii [2] or caratuberside E from Caralluma tuberculata [3]. Tested at 100µM, marnieranosides A and B stimulated the secretion of GLP-1 in STC-1 cells by 130%, as well as P57. Our results suggest that the potential antidiabetic properties observed with some Asclepiadoideae could be partly mediated via the GLP-1 pathway, and that glycosylated pregnanes could be responsible for these effects.
