Chemical profiling of Byrsonima species by HPLC-DAD-MS/MS and the evaluation of antioxidant, anti-inflammatory and cytotoxic activities

The use of analytical techniques, metabolomic tools and dereplication have increased in the field of natural products, making it possible to distinguish new compounds from those already described in complex matrixes [1], and have been shown to be very useful for identification of the all metabolites of plant species. Byrsonima is a genus occurring in the most part of Brazil, and it is known for its antimicrobial activity. This genus is the largest of the Malpighiaceae taxon, and comprises species of medicinal importance, ornamental plants, and producers of edible fruits [2]. The fingerprinting of secondary metabolites of four different Byrsonima methanolic extracts (B. intermedia, B. coccolobifolia, B. verbascifolia and B. sericea) were obtained by HPLC-DAD. The evaluation of in vitro antioxidant activity by DPPH·, ABTS+·, and pyranine-based assays, anti-inflammatory activity by NO production assay and cytotoxic activity showed the pharmacological potential of these plant species. Byrsonima sericea DC extract seems to be the most antioxidant in all assays (EC50 9.4 µg/mL for DPPH·, 2.99 for ABTS+· and 1.60 for ROO· scavenging), and the most potent to inhibit NO production in the presence of LPS (63% at 12.5 µg/mL). In all extracts no cytotoxic effect was observed. A micro scale bio-guided fractionation of the crude extracts was done and the main compounds in the most active fractions were tentatively identified by HPLC-DAD-ESI-MS/MS and dereplication approach, using the molecular masses, together with the fragmentation patterns, UV spectra, and data from literature [3,4]. About 20 compounds were identified, including several flavonoid glycosides, mainly derived from quercetin, gallic acid derivatives, galloylquinic acids and proanthocyanidins, which were reported to be active.
