Formulation and evaluation of paeoniflorin clubbed lipospheres for treatment of rheumatoid arthritis

Rheumatoid arthritis is a systemic autoimmune disorder. Chronic inflammation and oxidative stress have been reported to be important causes of arthritis. Paeoniflorin, an active glycoside constituent obtained from plant Paeonia lactiflora, possesses analgesic, anti-inflammatory, antioxidant, immunomodulatory activity and inhibits joint destruction. Paeoniflorin clubbed with in lipospheres for treatment of rheumatoid arthritis. The nine formulations (LP-1-LP9) of Paeoniflorin-loaded polymeric lipospheres were characterized for morphological parameters, PDI, entrapment efficiency, in vitro drug release. The size and PDI of LP-5 and LP-6 were found to be 206 ± 4nm, 0.30 ± 0.10 and 228 ± 6nm, 0.24 ± 0.4, with entrapment efficiency of 88% and 91%, respectively. Analgesic and in vivo activity were evaluated in collagen-induced arthritis rat models. These findings were supported by the histopathological characteristics of synovium sections. Observation revealed inhibition in proliferation of synoviocytes and production of cartilage destruction enzyme MMP-1, MMP-3. These result obtained assures that paeoniflorin-lipospheres could be easily manifested for treating root cause of arthritis.
