Karlotoxins, a group of dinoflagellate derived polyketides, have attracted much attention as potential therapeutic compounds due to their potent ichthyotoxic, cytotoxic and hemolytic activities related to their special structural features. Here we describe synthesis of major fragments toward the stereoselective total synthesis of karlotoxin 5, a recently isolated karlotoxin 2 congener from the marine dinoflagellate Karlodinium veneficum
