Orthogonal Determination of Standardization Markers in Accelerated Solvent Extractions of Spent Hops (Humulus lupulus L.)

Quantitation of phytochemical markers, either bioactive or not, is an essential step for the characterization and quality control of botanical extracts, contributing ultimately to their standardization. In the present study, spent hops were extracted by ASE with a DIONEX Model 350. As a quality control measure, the chemical markers profiles in spent hops extracts were compared, and the multi-target quantitation of characteristic chalcone (xanthohumol [1]) and flavanones (isoxanthohumol [2], 8-prenylnaringenin [3], and 6-prenylnaringenin [4]) was performed with a Shimadzu Nexera UHPLC equipped with a Waters Acquity UPLC ® BEH C18 (2.1 × 5.0 mm, 1.7 µm) column and using PDA and fluorescence as detection modes. The quantitative method, validated in terms of precision, accuracy, and repeatability, allows a rapid quality evaluation of various hops extracts. In order to build a standardization method for a clinical hops extract, an orthogonal validation using 1D and 2D 1H NMR will be presented.
