Rapid scaleup of high performance countercurrent chromatography from bench to kilogram

Countercurrent chromatography (CCC) has found broad application for natural product purification at the discovery scale but one limitation to the practical use of the technique has been rapid and predictable scaleup. This limitation has been overcome using HPCCC (HPCCC) instrumentation. As an example, method development and the kilogram scale purification of two semi-synthetic spinosyns, spinetoram-J (1) and -L (2), at the kilogram scale will be described.
