Effect of Oral Tetracycline on Iron Absorption from Iron(III)-hydroxide Polymaltose Complex in Patients with Iron Deficiency Anemia

Abstract

The study was carried out as an open-label, laboratory-blind, single-dose, randomized, two-period crossover, isotope efficacy study. Twenty-two patients with iron-deficiency anemia were enrolled in the study. The study consisted of two treatment phases of 15 days each, including blood sample measurements for Fe-59 activity. The two treatments were given orally. Treatment A was Fe-59 labeled iron(III)-hydroxide polymaltose complex (IPC, Maltofer©) equivalent to 100 mg elemental iron given orally. Treatment B consisted of Fe-59 labeled IPC complex equivalent to 100 mg elemental iron and 500 mg tetracycline HCl (CAS 64-75-5) given orally.

No differences between the two treatment groups with regard to the erythrocyte iron uptake were found, and thus IPC can be used with tetracycline, if necessary.

Zusammenfassung

Einfluss von oral verabreichtem Tetracyclin auf die Eisen-Absorption aus Eisen(III)-hydroxid- Polymaltose- Komplex bei Patienten mit Eisenmangelanämie / Randomisierte kontrollierte Isotop-Studie an einem Zentrum

An Patienten mit Eisenmangelanämie wurde der Einfluss von Tetracyclin HCl (CAS 64-75-5) auf die Absorption von Eisen(III) -hydroxid- Polymaltose- Komplex (IPC, Maltofer©) untersucht. Die Studie wurde als offene, labor-verblindete monozentrische randomisierte Zwei-Pha-sen-Cross-over-Studie mit einmaliger Gabe von Test- bzw. Referenzmedikation durchgeführt. 22 Patienten mit Eisenmangelanämie beendeten die Studie mit 2 Behandlungsphasen von je 15 Tagen Dauer, getrennt durch eine Auswaschperiode von 21 bis 28 Tagen. Beide Behandlungen wurden oral verabreicht. Behandlung A bestand aus Fe-59-markiertem IPC mit einer Dosis von 100 mg Eisen. Behandlung B bestand aus Fe-59-markier-tem IPC, entsprechend einer Dosis von 100 mg Eisen und 500 mg Tetracyclin HCl.

Es konnten keine Unterschiede zwischen den beiden Behandlungsgruppen im Hinblick auf die Fe-59-Aufnahme in die Erythrozyten gefunden werden, was bedeutet, dass IPC, falls nötig, mit Tetracyclin verabreicht werden kann.
