Simultaneous Quantification of Eight Active Compounds of Ojeoksan by HPLC Coupled with Diode Array Detector

A systematic high performance liquid chromatographic with diode array detector (HPLC-DAD) method was developed for rapidly simultaneous determination of eight marker components, paeoniflorin, 6-gingerol, decursin, glycyrrhzin, cinnamic acid, hesperidin, magnolol and poncirin in a traditional herbal medicine, Ojeoksan. To establish HPLC-DAD method, DIONEX C18 column (5µm, 120 Å, 4.6mm×150mm) was used with gradient elution system of water and methanol. UV wavelength was set at 230nm, 254nm and 280nm. Linearity, recovery, and precision tests were carried out for validation of this method. A calibration curve of standard components showed excellent linearity (R2>0.9922). Limit of detection (LOD) and limit of quantification (LOQ) were ranged from 0.01 to 0.11µg/ml and 0.03 to 0.34µg/ml, respectively. The relative standard deviations (RSDs) of data of the intra-day and inter-day experiments were less than 2.15% and 2.60%, respectively. The accuracy of recovery test results were ranged from 94.27 to 107.68% with RSD values of 0.15–3.61%. The results of validation suggested that this method was very accurate and stabilized for quality of control of Ojeoksan.
