Untractable Hypoglycemia in an Infant: Studies on Pancreas Insulin and Glucagon

Abstract
Insulin and glucagon were extracted from the pancreas of a 2-month old child with untractable hypoglycemia. High contents of insulin (680 µg/g of wet weight) and of glucagon (84 µg/g of wet weight) were found. After gel filtration, the different fractions were analysed by radioimmunosassy, (RIA) and radioreceptorassay (RRA) for insulin and glucagon. The proinsulin fraction represented 5.4% and 1.6% of the total insulin in the RIA and the RRA, respectively. Five percent of the glucagon was found as a larger size component. The insulin and glucagon fractions behaved identically to the reference hormones in both the RIA and the RRA; the glucagon fraction was as potent as the reference glucagon in stimulating adenylate cyclase in liver membranes.
