Benzyladenine and kinetin influence on skin fibroblasts proliferation and morphology

Benzyladenine and kinetin are cytokinins – adenine analogs that act as hormones to control many processes in plants [1]. We tested their influence on human skin fibroblasts. The fibroblasts cell line, derived from a patient was maintained in DMEM medium supplemented with 10% fetal bovine serum at 37°C in a humified atmosphere of 5% CO2 in air. Suspensions of cells (1×105 cells/ml) in 2ml of culture medium were incubated with or without the test compounds in tissue culture plates 6-well. We tested cell proliferation at 10-3÷10-8 M concentration of kinetin and 10-3÷10-8 M benzyladenine. Cell numbers were counted in haemacytometer acc to Bürker, double ruling by colouring dead cells with methylene blue. We also observed changes in cells morphology. Kinetin was more efficient than benzyladenine. Benzyladenine stimulated cells proliferation at 10-7M, and kinetin at 10-5÷10-6M. 10-3 M concentration of benzyladenine and kinetin acted lethally. These data suggest that both kinetin and benzyladenine stimulate cells proliferation and influence on their morphology. Further experimentation is necessary to understand the mechanisms of benzyladenine and kinetin action in cell and their influence on protein biosynthesis, especially collagen and connected enzymes activity.
