Analysis of gene and protein expression of p53 and Bcl-2 in human breast cancer T47D cells treated with extracts of Astrodaucus persicus (Boiss.) Drude
The effects of Astrodaucus persicus, an Iranian species of family of Umbelliferae, on p53 and Bcl-2 mRNA and protein expression in human breast cancer T47D cells were studied. The mRNA levels of p53 and Bcl-2 were analyzed in T47D cell line using RT-PCR. The p53 and Bcl-2 proteins expression in this cell line were also analyzed using immunocytochemistry (ICC) technique. In comparison to RPMI control, the p53 gene expression increased in presence of aerial and root extracts. On the other hand, the Bcl-2 gene expression significantly increased in presence of aerial extract but decreased in presence of root extract. Both extracts also showed decrease in p53 and Bcl-2 protein expression in comparison to control RPMI. Iran has unique plant varieties yet to be studied for anticancer components. These data indicates potential anticancer activity of A. persicus extracts and its possible molecular mechanisms of action on breast cancer cell proliferation.
