Anti-inflammatory and antioxidant activities of Haplopappus taeda extracts

Haplopappus taeda (“Bailahuén“), is a native medicinal Haplopappus (Asteraceae) species included within a“Bailahuén complex“, frequently used in folk medicine for liver ailments[1]. The aim of this work was to show that this pharmacological effect could be associated with anti-inflammatory properties of secondary metabolites present in the leaves. To this purpose, the ethanolic extract of fresh plant (Ht) was submitted to assay-guided chromatographic fractionation to give three main active compounds that were identified by spectroscopic methods. Anti-inflammatory effect was evaluated in vivo using the AA-induced acute ear oedema [2] assay (20µL, 3.6×10-4M/ear) along with in vitro assays (free radical scavenging with DPPH [3], Xanthine oxidase inhibition and superoxide anion scavenging [4]). The results show a high anti-inflammatory activity of Ht (72.7%±23.1) that derives mainly from the monoterpene taedol (45.7%±11.2), the clerodane diterpene ent-5α-18-acetoxy-cis-cleroda-3,13 E-dien-15-oic acid (30.1%±12) and the flavanone sakuranetin (34.3%±11.5). Under the same experimental conditions, nimesulide displayed a maximum of 48.8%±4 topical anti-inflamatory effect. Significance of results was calculated using Wilcoxon's non parametric method for independent data. Effects calculated are relative to the mean value±S.E.M.
