Studies on standardized extracts of Schisandra chinensis

Fructus Schisandrae, originated from the dried ripening fruits of Schisandra chinensis (Turcz.) Baill. (Magnoliaceae) is applied in oriental medicine and 'Shen Nong's Herbal Classic' ranked it into the high grade. Standardized Extracts of Traditional Chinese Medicine refers to an extracted and processed product of Chinese crude drug with relatively definite medical substance and stringent quality specification. In this study, a relatively all-round research of two kinds of Galenical-type extracts, one kind of Semi-purified extracts and three kinds of Selective phytochemical extracts of Fructus Schisandrae had been carried out. The preparation procedure, chemical composition, and quality evaluation were briefly dealt with. Total count assaying methodology of three types of principal chemical constituents in 3 kinds of extract were screened and ascertained and these assaying results can mostly reflect the realities of content determination. Furthermore; the chemical constituents were actually enriched in diverse extent to meet different requirements. Chemical reference substances in lignan extract were isolated by means of silica gel column chromatography, preparation TLC, Sephadex column chromatography, HPLC preparation chromatography and recrystallization, their structures were identified by NMR data and comparison with known compounds. The qualities of four Standardized Extracts from different habitat and in different collecting time were evaluated, including total content assaying of three types of chemical composition and content assaying of one lignan. The method of HPLC fingerprint spectrum on the studies of lignan extract is applied for the first time and quality control pattern of the extracts with fingerprint was established. It was set up that the full-scale quality evaluation on lignan extract from different habitat and in different collecting time. The rationality and correctness were authenticated by cluster analysis method. The contents of their moisture and ash were determined and the content of heavy metals was also analyzed with the method of atomic absorption spectrophotometry and chromatometry. All the above study certified the standard extract satisfied the need of phytoequivalence in medicine usage better than crude drug.
