Abstract

In the search for potent and selective 5-lipoxygenase-inhibitors, extracts of medicinal plants were screened for their potency of inhibiting the formation of inflammatory acting products of arachidonic acid metabolism using a porcine-leukocytes test system. The coumestan derivative wedelolactone isolated from Eclipta alba (L.) Hassk. and Wedelia calendulacea Less. has been found as a potent and selective 5-lipoxygenase-inhibitor with an IC50 of 2,5 µMol. As found by chemoluminescence, wedelolactone inhibits 5-lipoxygenase by an oxygen radical scavenger mechanism.
